The choke plunger o ring. It will unscrew with wrench from the left carb. -

Get a compression tester. First pull the plug caps and trim 1/4 in off and reinstall -
I responded but it didn't send for some reason just resent it. Let me know if you don't get it

That missing choke tube is part of your problem. Replace it and try again. Clean your carbs and install that tube and try to run it -

I tell you what. You said it will start if you put fuel in the cylinder. Get it started, choke it(full choke)and see if the fuel flows between the carbs. Try this first

I would say that there is a clogged up passage on the choke circuit. Loosen the screws on the carb bowls and make sure (again) that fuel will flow out of the bowls. If it does then that left carb is still dirty

It won't be a lot but when you kick it with the choke on it will kinda mist thru. Look real close. Kick it ten ten times with the choke on then remove tube and see if there is evidence of fuel

When you kick it you should see fuel going from carb to carb in the choke tube if the hose is clear. Check that out.
