First of all as my first post i would like to say hi from australia.I have a TDR250 which for those that dont know the model is a road trail bike with a TZR250 engine.I would like some advice on the 535cc Cheetah big bore kit.I am thinking of getting one of these kits for an RZ350 bottom end for the TDR.What are you thoughts on this?Any hints/set up tips would be greatly appreciated....heres a pic of the bike

Welcome...a 535cc kit is not a bolt on and go kit...there is some modifications that will need to be done to your cases to fit the crank. if you haven't already do search on this and you will find alot of info..

A 421 kit however is basically bolt on. You may consider that. With the right carbs/pipes and tuning a 421 will make about 90Hp on gas I believe. Probably more if you had further port work done.
